SS-31 (also known as Elamipretide, Bendavia, or MTP-131) is a first-in-class synthetic tetrapeptide that selectively targets the mitochondria. It belongs to the Szeto-Schiller (SS) family of peptides, characterized by their ability to cross the blood-brain barrier and penetrate cell membranes without the need for specific transporters.

Primary Mechanism of Action

The unique functionality of SS-31 stems from its interaction with Cardiolipin, a phospholipid found exclusively on the inner mitochondrial membrane.

 

  1. Cardiolipin Stabilization: SS-31 binds to cardiolipin, preventing it from undergoing oxidative damage. This stabilizes the mitochondrial cristae structure, which is essential for energy production.

     

  2. ATP Enhancement: By optimizing the electron transport chain (ETC) and improving the organization of mitochondrial "supercomplexes," it facilitates more efficient ATP (cellular energy) synthesis.

     

  3. ROS Scavenging: It significantly reduces the production of Reactive Oxygen Species (ROS), protecting cells from oxidative stress and preventing mitochondrial swelling.

     

  4. Inhibition of mPTP: It helps inhibit the opening of the mitochondrial permeability transition pore (mPTP), a key event that leads to cell death (apoptosis) during times of cellular stress.
